By acting through both GIP and GLP-1 receptor pathways, tirzepatide targets multiple pathways involved in appetite and metabolic regulation.

The SURMOUNT-1 phase 3 trial evaluated tirzepatide in 2,539 adults with overweight or obesity over 72 weeks and demonstrated substantial reductions in body weight across the studied treatment groups.


Retatrutide

Retatrutide is an investigational triple-receptor agonist with activity at:

  • GIP receptors
  • GLP-1 receptors
  • Glucagon receptors

A phase 2 randomized trial evaluated retatrutide for 48 weeks in adults with obesity and demonstrated substantial reductions in body weight across the studied treatment groups.

Retatrutide remains investigational and is not FDA-approved. Its regulatory status and availability differ from FDA-approved prescription medications.
